摘 要:该研究以铁皮石斛水提液为主料,添加人参、黄精、枸杞等辅料,开发一款能够改善药物性肝损伤的复合饮品,并深入探究其对药物性肝损伤小鼠的调节功能。通过感官评定结合模糊数学法,综合单因素、Plackett-Burman试验以及Box-Behnken响应面试验,确定最佳配方:铁皮石斛水提液添加量3.44%、人参水提液添加量0.81%、黄精水提液添加量0.12%、枸杞水提液添加量1.0%、冰糖液添加量4.26%,此配方下产品感官评分最优。动物试验中,通过腹腔注射环磷酰胺成功建立小鼠肝脏损伤模型,并连续给药治疗28 d。结果表明,与模型组相比,复合饮料高剂量组的小鼠肝脏指数显著降低(P<0.05),血清中AST、ALT水平以及肝脏氧化指标MDA含量均极显著下降(P<0.01),SOD水平则极显著上升(P<0.01)。肝脏病理学形态观察也有力佐证了该复合饮料对药物性肝损伤具有高效的改善与修复能力。

Preparation Process of Dendrobium officinale Compound Beverage and Its Regulatory Effect on Liver Injury in Mice Induced by Cyclophosphamide

Abstract:This study developed a compound beverage with Dendrobium officinale aqueous extract as the main ingredient and ginseng,Polygonatum odoratum,and Lycium barbarum as auxiliary ingredients to alleviate drug induced liver injury (DILI). The regulatory function of the compound beverage on DILI in mice was also deeply explored. Through sensory evaluation combined with fuzzy mathematics method,by integrating tests of single factor,Plackett-Burman,and Box-Behnken response surface,the adding amounts of optimal formula were determined:3.44% Dendrobium officinale aqueous extract,0.81% ginseng aqueous extract,0.12% Polygonatum odoratum aqueous extract,1.0% Lycium barbarum aqueous extract,and 4.26% rock sugar solution. The sensory score of the product was the best under this formula. In the animal experiment,a mouse liver injury model was successfully established by intraperitoneal injection of cyclophosphamide followed by 28 d of continuous administration of the compound beverage. The results showed that compared with the model group,the liver index of the high dose compound beverage group was significantly reduced (P<0.05),and the levels of AST,ALT in serum and liver oxidation index MDA content in liver were all extremely decreased (P<0.01),while the level of SOD was extremely increased (P<0.01). The pathological morphology observation of the liver also strongly supported that the compound beverage had an efficient alleviation and repair ability for DILI.
